Cell Protection Technology

Objectives

The aim of JRG Cell Protection Technology is to develop bio-formulation and protection strategies for stem cells as well as 3D native and engineered tissues for safe transport and long-term storage to facilitate their availability for future medical applications. Our research findings will lead to an extended shelf life of donor cells, tissues and organs until transplantation.

Research Focus

For the successful realization of stem cell-based therapeutic approaches in regenerative medicine, there is an urgent need for safe and effective enabling technologies for long-term storage of biological material, such that they can be acquired on demand, without loss of quality. Cryopreservation is a technology that allows the long term preservation of various cell types at extremely low temperatures. However, despite the popularity of cryopreservation in industrial and academic applications, effects of conventional cryopreservation procedures on subcellular properties such as epigenetic mechanisms have not yet been properly characterized. Furthermore there is a need for alternative cryoprotective strategies, especially since most widely used cryoprotective agents such as DMSO have been shown to have time-, temperature- and concentration-dependent cytotoxic effects. New strategies include reduction of DMSO concentration by application of sugars, antifreeze and ice binding proteins along with cell electroporation for efficient CPA delivery. Moreover, control over ice nucleation temperature and thawing rate could significantly improve quality of cryopreserved cells and tissues.

Another challenge in the field of low temperature biology is the storage of 3D tissue samples. Current technologies make it difficult to perform functional studies on 3D tissue-derived living cells, after thawing. Recent advances in tissue engineering such as cell and organ transplantation and development of protection methods like embedding or encapsulation offer promising options for the storage and transport of stem cells and therapeutic products.
